Immune Profiling Nanobody Panel
The Immune Profiling Nanobody Panel is a multiplexed binder set of oligonucleotide-conjugated anti-human VHH nanobodies (~15 kDa single-domain antibodies) for simultaneous immune cell surface marker detection on the MACS Matchmaker focal molography platform.
The panel covers eight well-established leukocyte surface markers:
- CD45: pan-leukocyte (all hematopoietic cells)
- CD3ε: pan-T cell
- CD4: helper T cells
- CD8: cytotoxic T cells
- CD19: B cells
- CD56: NK cells
- CD14: monocytes
- CD7: T/NK lineage
This marker combination resolves the core immune cell classes: the T-cell compartment with helper and cytotoxic subset discrimination (CD3ε/CD4/CD8), B-cell identification (CD19), NK-cell and lymphocyte-lineage profiling (CD56/CD7), monocyte detection (CD14), and pan-leukocyte gating (CD45).
Each nanobody is conjugated to a sequence-specific oligonucleotide for DNA-directed immobilization (DDI) onto complementary capture sequences on the sensor chip surface. The nanobody format offers key advantages over conventional IgG antibodies: compact size (~15 kDa) for higher packing density on the mologram ridges, a single binding domain that eliminates Fc-mediated non-specific interactions, and a defined conjugation geometry for uniform surface orientation.
The panel runs on the Sensor [Oligo|Oligo] 8-plex for simultaneous profiling of all eight markers in a single measurement, and is compatible with complex biological matrices including whole blood, PBMCs, and serum-containing media.
Each nanobody adapter is supplied pre-conjugated to the oligonucleotide sequence of choice (Seq ID 00 to Seq ID 16), so a distinct sequence can be assigned to each binder for flexible multiplexing on the sensor chip.
