Oligonucleotide Conjugation Kit, AminoLink

The Oligonucleotide Conjugation Kit, AminoLink enables the easy and efficient generation of protein-oligonucleotide conjugates in less than 4 hours. This kit is optimized for the conjugation of 100 µg of protein (15 – 300 kDa) to proprietary ssDNA oligonucleotides (see complimentary Oligo Adaptor TCO (https://shop.lino-biotech.com/products/oligo-adapter-for-amine-kit) ).

The conjugation of protein to oligonucleotides is achieved in several consecutive steps: 

2. Starting with the chemical activation of the protein with the help of a bifunctional linker. The linker reacts with an amine (NH2) group on the protein and activates it with a bioorthogonal methyltetrazine (MeTz) moiety.  
4. Once modified, the protein can be conjugated to DNA-oligonucleotides containing a trans-cyclooctene (TCO) group.
6. After ligation, excessive oligonucleotides are removed through washing and purification steps.

The conjugation protocol provides flexibility in the protein-to-oligonucleotide conjugation ratio and allows fine-tuning the ratio from 1:1 up to 1:5, depending on the downstream assay.
